Right away I can see that Ollie Matson is maybe the biggest snub here. Traded for NINE players (included were some draft picks) to the Rams in the late 50's. It was the Herschel Walker trade before the Herschel Walker trade (though it failed to set the team up for years to come like the Walker trade did for Dallas).
If Night Train wasn't on the Detroit roster, he would have go be here also. I'd place him on the Lions, though most of his best years were spent with the Cardinals.
I think if we're looking at KR, Vai is about all you need, but remember Terry Metcalf also was a great kick returner (as well as Ollie Matson)...he could've been the other (save a roster spot also).
Johnny Roland and Jim Otis would be candidates for another RB spot if necessary, and Calais Campbell at DE.
Fun fact - years ago I set up a bunch of these all time teams on Madden and ran a tournament for a few weeks...the winner of the tourney...the Cardinals. Over Detroit. Go figure.
They're also one of the oldest continually run franchises in the league (which gets lost sometimes due to the moves - Chicago, St. Louis, Arizona - and their futility).
I will say - it's a tough secondary to top with three HOFers (Wehrli, L. Wilson, Williams)and another that could be one day (Adrian Wilson).

Kurt Warner could be on here. I know he did most of his career with the Rams, but he is 4th all time Passing for the Cardinals and led them to their only Super Bowl appearance.
